{"id":20239,"date":"2021-07-07T16:46:47","date_gmt":"2021-07-07T15:46:47","guid":{"rendered":"https:\/\/photonics.laser2000.co.uk\/?p=20239"},"modified":"2021-07-07T16:46:47","modified_gmt":"2021-07-07T15:46:47","slug":"burnt-out-filter-for-fluorescence-microscopy","status":"publish","type":"post","link":"https:\/\/stagingphotonics.laser2000.co.uk\/?p=20239","title":{"rendered":"Burnt out filter for fluorescence microscopy"},"content":{"rendered":"\n<h4 class=\"wp-block-heading\" id=\"h-how-to-deal-with-metal-halide-lamp-burning-out-your-filter-for-fluorescence-microscopy\">How to deal with metal halide lamp burning out your filter for fluorescence microscopy.<\/h4>\n\n\n\n<p id=\"Contents\">This short case study covers one of our customer&#8217;s challenges with a burnt out fluorescence microscopy filter, what the solution was and the result.&nbsp;<\/p>\n\n\n\n<h3 class=\"has-text-align-center wp-block-heading\" id=\"h-the-challenge\"><strong>The Challenge<\/strong><\/h3>\n\n\n\n<p>Researchers at the University of Cambridge were trying to image fluorescence samples on their Olympus IX51 microscope, when they noticed that they had to keep increasing the exposure time of their experiment in order to get the same quality image as they had previously acquired. They asked us to help get to the root of the problem.<\/p>\n\n\n\n<p>At Laser 2000 we believe it is of the upmost importance that our customers are satisfied and have the equipment that they need, in&nbsp;<em>full<\/em>&nbsp;working order. This includes coming to&nbsp;<em>you<\/em>.<\/p>\n\n\n\n<p>On the visit to the laboratory in Cambridge, we discovered that one of the filters in the microscope had burnt out. The cause of this was highly likely to be the fact that the previous light source for the microscope was a metal halide lamp. Even though this was changed to an LED light source some time ago, the filters were never changed. The high Ultra-Violet component from the metal halide lamp most likely burnt out the filter over time, which then prevented the LED light from being transmitted, reducing the DAPI excitation and increasing experiment runtime.<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-large\"><img decoding=\"async\" src=\"https:\/\/stagingphotonics.laser2000.co.uk\/wp-content\/uploads\/2021\/07\/Screenshot-2021-07-07-105114.png\" alt=\"\" class=\"wp-image-20241\" \/><\/figure><\/div>\n\n\n\n<h3 class=\"has-text-align-center wp-block-heading\" id=\"h-the-solution\"><strong>The solution<\/strong><\/h3>\n\n\n\n<p>After identifying the burn out issue, we replaced the filter set&nbsp;<a href=\"\/products\/light-delivery-and-control\/microscopy-filters\/filter-sets-cubes\/single-band-microscopy-sets-filter-sets-cubes-microscopy-filters\/brightline-single-band-filter-set-dapi-and-similar-fluorophores-illuminated-with-led-based-light-engines\/\" target=\"_blank\" rel=\"noreferrer noopener\">with one from Semrock<\/a>&nbsp;\u2013 the pioneers of the fluorescence filter world, known for their high-quality products. The filter set was optimised for LED-based light sources as the spectral output of these light sources differs from that of metal halides. By installing hard coated Semrock filters that are optimised for LED-based light sources, we solved the original burn out issue as well as optimising the experiment.
